As an inhibitor of TORC1, an enzyme that regulates cell growth and metabolism, rapamycin is known to hinder cell growth in post-transplant and cancer patients by suppressing the immune response. However, it can also significantly extend longevity by interfering with the activity of the protein called mammalian TOR, or mTOR.
When administered lifelong, rapamycin has been found to delay aging and extend the lifespan of numerous animal models, decelerating cognitive decline, cancers, cardiovascular aging, and immune impairment. In more current human studies, short courses of rapamycin have been shown to improve immune responses in older people. This is why subsequent research is being conducted to determine if the drug can lead to medicines that mitigate aging-related disorders, such as Alzheimer’s disease, cancer, and heart failure.
Currently, rapamycin has not been approved by the FDA as a life extension drug.
